A LuxR/lux-box circuit expresses PLA1 in response to AHL, lysing dye-loaded storage vesicles inside GUVs and releasing CPRG for cleavage by beta-galactosidase, giving a visible color change that distinguishes AHL-induced from non-induced conditions within about an hour.

A TetR-repressed, anhydrotetracycline-inducible deGFP cell-free reaction is encapsulated in giant unilamellar vesicles by inverted emulsion transfer, and its dose-response is read by fluorescence microscopy against a matched unencapsulated plate reader control.

TetR represses the pT7-tetO-plamGFP cell-free reporter to near background at 500 and 1000 nM, and a seven-point anhydrotetracycline (aTc) titration relieves that repression fully by 5 uM; the amount of TetR sets where the induction threshold sits rather than whether the switch works.

Phospholipase A1 (PLA1), expressed in a cell-free reaction and placed under TetR/anhydrotetracycline control, is delivered to the outside of giant unilamellar vesicles loaded with beta-galactosidase to ask whether externally supplied PLA1 can lyse the membrane, and whether that lysis can be triggered on demand.

Co-encapsulating a complete cell-free reaction with beta-galactosidase inside GUVs shows that protein expression itself, not phospholipase activity, drives loss of membrane containment; a follow-up experiment confirms the effect is graded and controlled by expression level via TetR/aTc, independent of which protein is made.

Whether vesicles stay stable and biochemically functional when embedded in a hydrogel, where they are mechanically confined and in contact with a crosslinked polymer network, read as T7-driven GFP expression in GUVs and as CPRG release from LUVs across three hydrogels and a solution control.

EsaR(D91G) repression of single- and dual-operator EsaO-mNG sensor constructs in Nucleus Cytosol, across five days: an EsaR titration at fixed sensor DNA, a sensor DNA titration at fixed EsaR, a repeat of that titration with EsaR pre-expressed overnight, a magnesium sweep with EsaR and DNA pre-incubated before the reaction, and an EsaR DNA template competition assay.

A synthetic-cell hydrogel that senses bacterial contamination through quorum-sensing-induced cell-free protein expression: POPC GUVs carrying LuxR-mNeonGreen lysate, embedded in agarose, fluorescing in response to 3-oxo-C6-HSL from purified standard or bacterial supernatant.

pH-gated PLA1 two-vesicle lysis cascade. PLA1-expressing GUVs co-incubated with CPRG-loaded LUVs across a pH 7.6 and a pH 6.3 arm, read by microscopy.
